A promising polymeric multilayered capsule targeting multiple myeloma with CPT and RB

Abstract (English)
Campitothacin is a promising topoisomerase inhibitor. However, its poor solubility and low stability limit its clinical applications. Converting Campitothacin from the carboxylate toxic form to the non-toxic lactone form while keeping its properties in cancer therapy is a promising strategy. In recent work, hyaluronic acid-functionalized multilayered polymeric capsules made using layer-by-layer techniques were used to deliver captothecin and rose bengal as a chemotherapy and photodynamic therapy approach targeting multiple myeloma cells overexpressing CD44. Polymeric multilayer capsules were fabricated on the surface structure of CaCO3 to induce (Chitosan/Polyacrylic acid)+(Polydopamine/Hyaluronic acid) layers after core removal. The result showed localized multilayered capsules in the perinuclear region after 24 h of cellular incubation. Following that, the measurement of general ROS and superoxide radicals was shown in both confocal microscopy images and flow cytometry, confirming ongoing apoptotic activation. Western blot showing the expression of cleaved caspase-3 in campitothacin-loaded multilayered capsules. It concluded that polymeric multilayered targeting nano therapies are considered a potential application for capitothacin and can protect healthy cells from side effects
